The difference is in the structure of the sentences.
DESPITE + NOUN
- Despite my best efforts, I couldnโt close the deal. (effort is a noun)
- Despite the fact that she had been warned twice, she made the same mistake. (fact is a noun)
- Despite being late, they let us participate in the game. (being is the noun form of be)

ALTHOUGH + SUBJECT and VERB
- Although I had done my best, I couldnโt close the deal. (I had done = subject and verb)
- Although she had been warned twice, she made the same mistake. (she had been = subject and verb)
- Although we were late, they let us participate in the game. (we were = subject and verb)
